Quantitative Enzyme-linked Immunosorbent Assay for the Estrogen-regulated Mr 24,000 Protein in Human Breast Tumors: Correlation with Estrogen and Progesterone Receptors1

چکیده
A competitive enzyme-linked immunoassay has been devel oped to quantitate the M, 24,000 estrogen-regulated protein (p24) in human breast tumors and tumor cell lines. The concen tration of monoclonal antibody used to coat polystyrene microtiter assay plates was critical in optimizing assay sensitivity versus precision and demonstrated marked differences between plate types. Analysis of 114 human breast tumor cytosols re vealed a 4-log range in p24 concentration with a median value of 1 fiQ/mg cytosol protein. The abundance of p24 in breast tumors in vivo is therefore similar to that observed previously in hormone-responsive breast tumor cells in vitro. Elevated expres sion of p24 in these specimens was found to correlate well with presence of estrogen and progesterone receptors (P = 0.0002 by Pearson x2 analysis) with the correlation coming primarily through estrogen receptor. This result was confirmed by immunoblot analysis of p24 in 76 additional tumor cytosols and suggests that p24 may be a valuable marker for differentiation in human breast cancer.

متن کاملStudy of estrogen receptor, progesterone receptor, and the estrogen-regulated Mr 24,000 protein in patients with carcinomas of the endometrium and cervix.
The presence of an estrogen-regulated protein with 24,000 molecular weight has been studied in 47 patients with endometrial carcinomas and in 29 patients with cervical carcinomas in order to correlate its presence with that of estrogen receptors (ERs) and progesterone receptors (PgRs). In the cytosol tumor samples the Mr 24,000 protein was detected by the Western blot technique using a monoclon...
متن کاملEnzyme-linked immunosorbent assay of c-erbB-2 oncoprotein in breast cancer.
Amplification or increased expression of the c-erbB-2 gene has previously been reported to be a prognostic marker for breast cancer. Gene amplification is usually measured by Southern blotting, whereas increased protein expression is usually detected by immunocytochemistry. We measured c-erbB-2 protein with an enzyme-linked immunosorbent assay (ELISA). High concentrations of oncoprotein were fo...

An estrogen-responsive murine Leydig cell tumor (M5480A) was examined for the presence of cross-reactive proteins to a monoclonal antibody directed against a Mr 24,000 estrogen-regulated protein in human breast cancer cells. Human breast tumor biopsies were used as controls for the cytosol preparations, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and Western blot conditions used ...

BACKGROUND Triple-negative breast cancer (TNBC) has negative expression of progesterone receptor (PR) and estrogen receptor (ER), and low expression of human epithelial growth factor receptor-2 (HER-2). This study aimed to investigate the expressional profile of cytokines in TNBC patients with significant expression of macrophages. MATERIAL/METHODS Immunohistochemical (IHC) S-P staining metho...
